Abstract: Harbinger2-1_ES belongs to a novel clade, Harbinger2, of Harbinger DNA transposons. This clade includes transposons present in protozoan (brown alga), fungi, choanoflagellate, and metazoans. Harbinger2-1_ES is a consensus sequence of a family of autonomous Harbinger transposons that were active in the red alga genome very recently. The consensus was derived from 3 copies >99% identical to it. The genome contains only two full-size copies of Harbinger2-1_ES; ther are flanked by the unusual TAA target site duplications (the same is for Harbinger2-3_TMe). In addition, the TWA target site duplications are perfectly defined for 8 copies of the non-autonomous Harbinger2-1N1_ES. TIRs are 16 bp long and contain 2 mismatches. HArbinger2-1_ES codes for the 350-aa TPase (3 exons).
